1. Development of robust, mass culture and cryo-preservation technologies for hPSC lines
(1) Synthetic, defined culture medium with low molecular weight chemical compounds Our initial results identified novel chemicals which have potentially allows the expansion of hPSCs efficiently. Utilizing Wnt and a small molecule, we have developed a novel and simple chemically defined xeno-free culture system.(2) Culture substrate and materials for large scale cell culture systems We have studied Hydogels which serve as the scaffold of hPSCs. For 3D mass culture of hPSCs we have succeeded to build robust culture procedure without no feeder suspension culture. (3) Closed and minimal risk cell culture system and 3D mass culture technologies We started to make the instruments on the basis of the specification. (4) Multi-parameter imaging technology for living cell in the culturing. The prototype of imaging system which can obtain multi-parameters without label has been designed and assembled.(5)Automated cryo-preservation technology for hPSCs We have evaluated the verification using the carboxylated polylysine (antifreeze polyamino acids).
