1. 研究開発の内容及び成果等 ヒトES細胞の安定な培養・保存技術の開発
研究者の手技の違いを排除し、一定条件での安定した培養操作を可能とする自動培養技術、ヒトES細胞の有用な性質を損なわずに安定培養が可能な、成分が明確かつ異種生物由来の成分を含まない培養液・培養基材の開発、凍結融解後の回収率が高く、ユーザーへの提供方法を考慮した凍結保存技術の開発を目的に、以下の技術開発を行なう。

 Integrated 3D culture system with some culture medium, substrates, and closed container will be developed.
This year, the development of technique which is feeder independent culture for human ES cells inside culture bag has been started. Along this development we have obtained some requirements for the culture bag and started test culture to obtain the data for three dimensional culture. On the development of automated culture instruments, we examined some tests using culture bag system, for adherent culture and decide on the specification of the instruments. We started to make the instruments on the basis of the specification. In the future, the condition of three dimensional cultures and the specification of automated culture instrument will be developed.
(4) Multi-parameter imaging technology for living cell in the culturing.
 To manufacture and provide “good” hESCs, continuous monitoring system for living
cultured hESCs are requested. This year, the prototype of imaging system which can obtain multi-parameters without label has been designed and assembled. This imaging system can measure solid profiles of ES cell colony such like optical thickness and adhesive characteristics, two dimensional profile such like perimeter or area of ES cell colony by transparent light imaging. Additionally, the imaging system can measure istribution of molecules which are fluorescent labeled, and also can find all of these
temporal and special change.
(5) Automated cryo-preservation technology for hESCs
 Skill-free, safe, and automatic cryopreservation technology will be developed.
This year, development of container for cryopreservation has been started.
The requirements of this container are closure form which can reduce risks of contamination.
