1) データベースの構築
デジタル標本データベースソースを作成するため、症例選択、各種染色を実施し、浜松ホトニクス社製バーチャルスライドスキャナーNanoZoomer 2.0-HTを用いて全スライド画像(Whole slide image, WSI)を取得した。

Data management and database construction: Whole slide images (WSI) of hepatocellular carcinoma (HCC) and non-neoplastic liver were obtained by digital slide scanner, NanoZoomer 2.0-HT (Hamamatsu Photonics). Region-of-interst (ROI) was selected from WSI at random. The information of hepatic cancer grade (Edmondson classification and differentiation) was added to ROI. 2. Development of digital image technology to histolgogical quantification of liver fibrosis: We develop the digital image technology to quantify the fibrotic area in elastica van Gieson stained sections, which discriminated collagen and elastic fibers from the other tissue components and assess liver fibrosis automatically. The fibrosis score calculated by a novel algorithm, correlated with the New Inuyama classification of liver fibrosis and data obtained by elastography. 3 Research and development of immunofluorescent digital slide technology to quantify protein expression in archival paraffin-embedded tissue sections: We have developed fluorescence-based, immunofluorescent quantification digital slides (IQD), a method widely applicable in routine practice. Sections were automatically stained with Leica BOND staining system (Leica Microsystems) and whole slide fluorescent images were obtained by digital slide scanner, NanoZoomer 2.0-HT (Hamamatsu Photonics). The signal sensitivity of IQD and DAB, both of which were stained with BOND under the same conditions were almost same. 4. Molecular marker assessment of HCC: Molecule X, which was a novel marker and drug target of HCC were assessed. We assessed mRNA expression of X in HCC cell lines by real time PCR and protein expression by western blotting. Using section of cell lines, we optimized algorithm for quantifying expression of X in histological section of HCC. Correlation between the amount of fluorescent dye and image signal was evaluated by correlative microscopy.
