英文要約We have developed a blood preparation method utilizing filtration to isolate cancer cells from peripheral blood. With the method, red blood cells and platelets in peripheral blood are effectively removed with the result that number of cells are reduced from 25 billion cells to 30 ~ 50 thousand cells per 5 mL blood (Preset goal is to reduce number of cells in 1 mL blood from 5 billion to 10 million). We succeeded in isolating cancer cells from pseudo CTC containing blood (5 mL of peripheral blood spiked with SKBR3 breast cancer cells) with the filtration method, with the result that rate of trapped cancer cells at the filter was 97 % (preset goal is over 90 %) and rate of collected cancer cells from the filter was 82 % (preset goal is over 80 %). We also demonstrated that a prototype system built in 2010 for cancer cell detection could detect single cancer cells from pseudo CTC containing blood (5 mL of peripheral blood spiked with SKBR3 breast cancer cells) with the result that detection rate was 80 % and repeatability rate was 6.0 %. Afterward, we build another prototype auto-system with functions of “blood preparation”, “fixing cells into the micropores”, and “labeling and detection of cancer cells”, and finished operation check of the system.
