英文要約The method which we developed utilizing filtration to isolate cancer cells
from peripheral blood has been revealed to be tend to lose small cells (H-69), and cells which are tend to alter cellular morphology (MDA-MB-231 and PC-14).The recovery rate of those cancer cells were between 1.8 % and 42.5 %. Thus, we have modified the protocol for CTC detection with a density gradientcentrifugation method (separating cancer cells from blood cells by differences of cell density) to isolate cancer cells from peripheral blood instead of the filtration method, with the result that recovery rate was 70 ~ 80 % regardless of type of cancer cells.
With the above protocol and a prototype system for cancer cell detection, we have detected cancer cells from pseudo CTC containing blood (3 mL of peripheral blood spiked with PC-9 lung cancer cells), with the result that detection rate was 87.0 % and repeatability rate was 11.0 %. We have also established a technique to sample single cells which are considered as CTC, and to find mutations in genome of the sampled single cells with DNA amplification by PCR technique.
