英文要約Title:Development of an evaluation system for blood samples (FY2010-FY2014)FY2012 Annual Report
An accurate inspection system detecting less than 100 CTCs in 10 ml peripheral blood has been developed. For performance confirmation of this system, we set to design and constructed the prototype composed of two processing units. One unit has the function of alignment and immunostaining of cells on a cell chip which has lots of microchamber-fabricated structure to retain a great number of cells. An automatically-controlled flow of the blood cell sample, prepared by density-gradient centrifugation to remove RBCs, allowed the chip hold in monolayer, and further we confirmed the efficiency of fluorescent multistaining cells on the chip in an automated way. Another unit functions as optical instruments for detecting rare cells. To identify CTCs among many WBCs, we employed the high speed scanning technology which enables rapid location of CTCs and subsequently high resolution images of CTC candidates were taken on fluorescent microscopy. By use of this prototype we could identify and analyze tumor cells spiked in whole blood sample.
