1. Research on Practical Application of Nano-imaging Technique for Cancer Tissues
As a result of staining study, grabbing fluorescent images and manual counting of fluorescent spots were achieved. From the next year, improvement of sensitivity including detection rate of the spots and exclusion of artifacts will be important.

2. Validation of the Clinical Values of This System
60 pathological samples from Tohoku University Hospital were stained and evaluated for fluorescent spots. It is found that matching ratio is 90% (55/61) compared with existing method (combination of DAB and FISH), if HER2 positive is defined more than or equal to two spots per cell. It is concluded that this one particle fluorescent spot measurement has high correlation with DAB or FISH. These result shows that the present 2 step diagnosis method using DAB and FISH could be replaced only by this method.
 Moreover we investigated the correlation between clinical result and one particle fluorescent spot score. Our one particle fluorescent spot method showed that one of the samples was HER2 positive, though it was diagnosed as “HER2 negative” by FISH method. This result might indicate our one particle fluorescent spot method is superior to FISH method for Herceptin response prognosis. To prove clinical value of one particle fluorescent spot measurement, selection and evaluation of various samples including for the patient having history of antibody drug treatment is important. The study will be continued for the next year in cooperation with Tohoku University Hospital.
