英文要約Title: Industrialization Research Project for Autonomous Regeneration Device (FY2010-FY2014) FY2012 Annual Report
Reconstruction of joints, based on autonomous regeneration of cartilages, would enable to promote generalization and industrialization of cartilage tissue engineering. In this project, we will establish an industrialization technology to apply the autonomous regeneration for reconstruction of joints. For that purpose, we have carried out research and development described below, this year. As industrialization of a hydrogel as a scaffold to realize autonomousc regeneration, we established the production condition of the hydrogel, PRG and accomplished the cartilage regeneration with approximately 1/100 cells compared to the conventional method in vivo, by applying the mixed gel consisting of PuraMatrix and PEG-Chitosan gel. As an early study of safety and efficacy for large animals, we carried on experiments on miniature pigs with only hydrogel and verified its safety. As industrialization of a culture module for construction of autonomic regeneration, we designed and produced a module, in which hollow fibers made of poly lactic acid cross-linked together. It contained two types of hollow fibers: one for the circulation of medium and the other for the excretion of waste matter. In addition, we developed a small module with an artificial bone and a hollow fiber made of poly lactic acid in it.
