英文要約Title: Development of Green Oil Production Process Using Cryophilic and Mesophilic Microalgae (FY2013-FY2015) FY2013 Annual Report

This project aims to develop an entire process for green oil production in which marine microalgal biomass will be converted to fuel materials with minimized energy consumption. Year-round production will be achieved using a well-studied mesophilic microalgal strain and newly isolated cryophilic strain in combination. Moreover life cycle analysis, in terms of energy balance, CO2 emission reduction, cost and stability of the established process will be investigated. The whole process is composed of 4 steps, (1) strain selection; (2) mass culture; (3) harvesting/dewatering; and (4) oil extraction. In this period, we mainly deal with (1), (2) and (4).
A mesophilic Fistulifera sp. strain JPCC DA0580 has been studied by our group as a promising oil producer with the advantages of high oil productivity and fast growth through spring to autumn seasons. To fulfill the gap at winter seasons, we screened cryophilic microalgae showing high oil content from our microalgal collection. As a result, a candidate strain JPCC CTDA0820 was demonstrated to show high potential in biofuel production. This strain exhibited a comparable oil accumulation with strain JPCC DA0580 even at low temperature (below 10 degree celsius). Algal biomass productivity reached 1.0 g/L in the modified f/2 medium.
Phylogenetic analyses revealed that strain JPCC CTDA0820 belonged to genus Mayamaea. Since Fistulifera sp. and Mayamaea sp. utilized in the present project are both marine diatom strains, it can be expected that both algal biomass can be processed by identical systems through oil production. Genomic information of strain JPCC CTDA0820 was collected, and a genetic manipulation technique was also established, thus metabolic engineering will be available in the future.
These laboratory scale experiments with promising results encouraged us to further investigate this strain under mass culture conditions. In this period, 10,000L low energy-type circle pond (8 units), oil accumulation pond (2 units), inoculation pond (2 units) and harvesting/dewatering facility were constructed. Mass cultivation of strain JPCC CTDA0820 in a 10,000L low energy-type circle pond was started from 17th of March, 2014. A countercurrent multi-step oil extraction technique was also proposed for these strains. Based on the culturing and oil extraction experiments, algal biomass and the extracted oil productivities in the circle pond were estimated to be 8.0 and 3.2 kg dried weight/day, respectively. These preliminary results in the mass culture experiments will be utilized for further optimization in the next research periods.
