The aims of our study are to establish effective metabolite biomarkers for quality evaluation of hPSCs and to develop a simple and quantitative method for measurement of the biomarkers using mass spectrometry (MS).
Results in this year
In this study, we tried to develop analytical method of primary metabolites (e.g., energy metabolism, amino acids, organic acids, sugars, and lipids) extracted from hESCs using gas chromatography (GC)-mass spectrometry (MS) and liquid-chromatography (LC)-MS. We also developed a method to evaluate changes of small compound in supernatant of hESC culture.
We observed 15 metabolites that showed a greater than two-hold difference with a p-value < 0.01 between undifferentiated hESCs and differentiated cells. It was also suggested that some lipids changed during differentiation. We further found a candidate metabolite that can discriminate difference of the passage numbers of hPSC culture.
