2. Development of quality evaluation and control for hPSC lines
(1) Development of profiling/evaluation hardware and reagents for quality control of hPSCs (human pluripotent stem cells, such as hES cells)

Progress of research in this year:
(1) Outline of the development:
Change on the generation and differentiation of human stem cells is an epigenetic process due to epigenetic modification of genes without changes in gene sequences. Novel quality assessment method to elucidate reliably epigenetic changes of stem cells as a means for its evaluation must be established.
Objectives of Genetein are (a) development a new Software/Hardware which is capable of automated epigenomic analysis for future quality assessment of hPSCs, and (b) Epigenomic analysis of hPSCs in different cultivation stages using the automated system .

(2) Achievements of the development
(2-1) Development of a new Software/Hardware
The new system has completed early this year and both short-term and long-term validation studies have been conducted including mechanical and wet trials using actual reagents. The results showed that the new system can perform well as expected.
(2-2) Epigenomic analysis of hPSCs
Using purified DNA from three different ES cells provided by Kyoto University(early stage, late stage and differentiated), automated MeDIP and h-MeDIP were conducted. The samples were analyzed by microarray. The data was analyzed by using algorithm to obtain DNA methylation and de-methylation patterns. Based on the results obtained, we have established a new method of distinguishing these hPSCs having different stages. Based on the new method, a patent was applied in January 2013.
The above achievement was presented in the World Stem Cell Summit held in Florida, USA in December, 2012. Also, joint press conference was held at Kyoto University on February
5th, 2013 under the title of "A new method of epigenome analysis by using an automated
epigenetic system" in collaboration with Genetein, Kyoto University, and NEDO.
