英文要約Research and Development of Next-generation Regenerative Technology / Research and development of in situ stem cell therapy using Muse cells (FY2010-FY2014) FY2013 Annual Report

This project clarifies the findings that form the base of in situ stem cell therapy using Muse cells, and aims to establish highly effective and safe next-generation cell therapy technology linked to in vivo autonomous tissue regeneration/repair and function recovery.

1.Research and development as to inducement of Muse cells into lesions

1-1.Comprehensive analysis of migratory factor:
We performed a search for candidate of migratory factor by referring to the results of an expression analysis or proteome analysis.

1-2.In vitro observation of Muse cell migration:
We continued the search for candidate of migratory factor. Also, we observed migration effect by using serum or damaged tissue in a disease model animal in vitro.

1-3.Observation of migration in model animal:
In model mice with liver degeneration, by tail intravenous administration of Muse cells, we have confirmed that Muse cells to be accumulated and integrated to injury site of liver where migratory factor is released.

2.Research, development and analysis on therapy by using migratory factor of Muse cells

2-1.Search for candidate substance:
We synthesized already identified small molecule compounds that bind specifically to the receptor, and examined their migration effect.

2-2.Study of therapeutic efficacy by administration and examination of conditions on model animals:
We have examined the therapeutic effect of migratory factor by administration and examination of conditions on model animals with skin defect.

2-3.Skin disease:
We performed a study, as a preparation of the implantation into the human, of function evaluation of the 3-D cultured skin which uses Muse cell-derived melanocytes.

2-4.Neurodegenerative disease:
We confirmed the therapeutic effect by administration of Muse cells or migratory factor to affected area of rat model with cerebral infarction.

2-5.Myocardial infarction:
We investigated the therapeutic effect by administration of Muse cells or migratory factor using a rabbit myocardial infarction model.

3. Research and development on device which engrafts Muse cells at lesion sites aiming for the verification of concept of cell therapy technology

3-1.Neurodegenerative disease:
We confirmed therapeutic effect by administration of Muse cell or migratory factor to affected area for neurodegenerative disease, and performed a study of the design and use of the device and a large or middle size animal model to use for this type of study.

3-2.Myocardial infarction:
For the purpose of verifying therapeutic effect of local administration of Muse cells or migratory factor to affected area or myocardial infarction of a large size animal model, we examined the test system and device design.

4.Survey and research for commercial use:
We coordinated the entire project, held the Development Committee and started negotiations with potential business partners for co-development.
