1) Construction of C5C6 co-fermenting microorganisms
The aim of this study is to construct the efficient ethanologenic yeast and to analyze the genetic information of the strain leading to potent xylose utilization, thermotolerance and fermentation-inhibitor tolerance in the simultaneous saccharification and co-fermentation (SSCF) process. In the FY2013, three xylitol-utilizing mutants of Saccharomyces cerevisiae were isolated. Genome analyses of these mutants were performed using the next-generation sequencer (NGS), mutations (SNPs and InDels) in those genomes were comprehensively solved, and we succeeded in identifying the causal gene mutations leading to efficient xylitol utilization of the mutants. We also sequenced a genome of thermotolerant yeast Kluyveromyces marxianus DMB1 to obtain genetic information related to thermotolerance by NGS. Furthermore, to develop standardized evaluation methods for fermentation ability of the genetically engineered yeast strains, several analytical methods for components in pretreated or saccharified biomass were examined.

2) Development of the SSCF process
In order to achieve target ethanol concentration of 5 w/v% in the SSCF process, more than 20 w/v% solid content biomass slurry must be handled in the reactor. Thus, at industrial scale, handling of the material is likely to be very difficult. Also, SSCF process is the , properties of biomass slurry (viscosity, particle size distribution, etc) changes as the reaction progresses. To obtain data related to slurry handling, therefore, to obtain data for slurry handling, 2m3 scale slurry handling apparatus was designed, and construction is started
