HMGB1, a nuclear protein, is thought to play roles in various inflammatory diseases and could be utilized as an important biomarker. Acute conditions such as sepsis, DIC (disseminated intravascular coagulation) and acute liver injury are amongst HMGB1-related inflammatory disease states that especially require rapid diagnosis and initiation of intervention. Due to the urgent nature of these diseases, an HMGB1 assay that (i) has short specimen preparation time, (ii) returns results rapidly and (iii) can be conducted near the patient (POCT; point-of-care testing). Shino-Test currently markets a research use only ELISA assay kit for HMGB1 which is used widely, however, the assay takes more than 22 hours which is far from the demands by the physicians in ER and ICU. This project aims to develop an assay system to meet such needs by combining unique proprietary technologies of Magnisense (France) and Shino-Test (Japan). Magnisense holds a sensitive electro-magneticism detection called MIAtek and Shino-Test has HMGB1-specific antibodies and related technologies.

The first phase of the development (proof-of-concept study), conducted mainly by Magnisense, is to explore applicability of Shino-Test’s antibodies and other know-hows to MIAtek technology. From Sept 2012 to March 2013, Magnisense explored various parameters and established conditions that enable one to quantitatively detect HMGB1 spiked in pooled serum, and the assay time is as little as 30 minutes.

Linearity, sensitivity and background signal level need to be improved for practical use of the assay, and the first phase will continue until the end of April 2013.
