英文要約Title:Development of an evaluation system for blood samples (FY2010-2014)FY2013 Annual Report
An accurate inspection system detecting less than 100 CTCs in 10 ml peripheral blood has been developed. For performance confirmation of this system, we set to design and constructed the upgraded prototype II composed of two processing units. One unit has the function of alignment and immunostaining of cells on a cell chip under an automatically-controlled flow conditions. By changing the pump construction this unit could obtain 4 times high throughput performance than before. Another unit functions as optical instruments for detecting rare cells. To identify CTCs among many WBCs, we added imaging function for acquirement of high resolution images of CTC candidates, to the high speed scanning technology. And then we started the construction of algorithm for distinguish between CTC and not CTC. By use of these technologies we have estimated the performance of our system using spiked tumor cells in whole blood. As a results, the recovery of tumor cells was about 70 percentage without depending on the expression level of EpCAM protein, especially MDA-MB-231 cell line, low expressing EpCAM, recovery was higher than CellSearch system. At the next step, we plan to perform a pilot study using breast cancer patient specimen.
