英文要約Owing to the system, we have detected cancer cells from pseudo CTC containing blood (3 mL of peripheral blood from health donor spiked with commercial SK-BR-3 breast cancer cells or commercial PC-9 lung cancer cells), with the result that the mean percentages of detected cells were - 70 % and mean coefficient of variation value was - 20 %, achieving our goal of this year.
In addition, we have established a method for collecting single CTCs and subsequent single-cell whole genome amplification.
Then we have fabricated a prototype system for research use, which consists of a sample-preparation apparatus and a detection apparatus, and have operated a clinical trial in which 3 mL of peripheral blood samples from each 15 metastatic breast cancer patients were measured.
As a result, we have detected CTCs from each blood samples. In addition, genomic analyses of amplified genome from collected single CTCs found mutations in a number of cancer related genes.
Meanwhile, two individual measurements of blood samples from healthy donors resulted in no false-positive detection.
