In this project, we investigated the development of a novel fermentation system for cellulosic ethanol production. During the fiscal year of 2014, we conducted a feasibility study of the following subjects.
1) Improvement in arming yeast
To implement the cell-surface display system into a practical yeast strain (i.e., diploid yeast), several enzymes including EGI, CBHI, and FAEI were expressed on the cell surface of Saccharomyces cerevisiae TJ14. The resulting yeast strains were used for ethanol fermentation of hydrothermally pretreated bagasse, in which an ethanol concentration over 4.5 wt% and yield of 85% were obtained successfully. The effective deletion of PHO13 in S. cerevisiae TJ14 has also been demonstrated.

2) Bench plant operation and evaluation for CBP
To consolidate the cellulosic ethanol production, we addressed the following subjects: (i) effect of pretreatment on enzymatic saccharification of bagasse; (ii) high cell-density cultivation of arming yeast; and (iii) liquefaction of pretreated bagasse followed by simultaneous saccharification and fermentation. As a result, we established the technical basis of CBP, which attains the cell density above 50 g/L, short-term liquefaction (within 5 hours), and ethanol titer of 50 g/L.

3) Business planning
To establish a business model, the following was investigated.
3.1 Marketing research
First of all, we are contacting with company A (US) regarding arming yeast, which was applied to the ethanol fermentation using the corn stover obtained from company A. From 7 to 11 December we visited Indonesia to attend the Indonesia-Japan business meeting on implementation of bio-refinery technologies held at IPB International Convention Centre in Bogor. We also visited LIPI, BPPT, and several companies who are producing ethanol, to which our new project was introduced. We found, through the research, that there are several candidates to collaborate with setting up the ethanol pilot plant.
3.2 Relocation of a pilot plant
We bought Hakutsuru sake brewery’s pilot fermenters in Kobe Shipping Yard of Mitsubishi Heavy Industries. We planned to move the pilot fermenters to our new research center in Nishi-Nakajima, Amagasaki, Hyogo.
3.3 Planning for scaling up a fermenter, cultivation of market, and fund procurement
Scaling up the ethanol fermenter up to 1,500L will be carried out in Japan and concerning to further scaling up we have to find a new business partner who has a raw material for ethanol production. We contacted with company B in Indonesia and she expressed future collaboration with us on a demonstration pilot plant of CBP process in one of her ethanol plants. The patents relating to the CBP process for ethanol production have already been applied to PCT, and re-applied to USA, Indonesia, India, Thailand, Malaysia, and Japan.
