英文要約The reproducibility of annual production of microalgae in open pond system (D=5 m) was confirmed using Mayamaea sp. JPCC CTDA0820 and Fistulifera solaris JPCC DA0580. Furthermore, oil productivity at outdoor culture was improved under a nutrition depletion condition. These performances will be examined in larger pond systems (D=20 m (310 m2、160 m3), D=40 m (1260 m2、640 m3)). Green oil could be directly extracted from algal culture by multi-stage extraction with n-hexane as a sole solvent. This result indicates that liquid-liquid extraction using WINTRAYR column was available for a low energy extraction process. Based on these results, total process of green oil production will be evaluated in the next year. Seawater intake facilities will be employed for the evaluation.
Furthermore, the draft genome sequence of cold-tolerant and oleaginous diatom, Mayamaea sp. JPCC CTDA0820 was determined, and several genes useful for metabolic engineering were identified. Also, gene knockout study revealed that genome editing technique using CRISPR/Cas9 system could be useful for oleaginous diatom F. solaris JPCC DA0580. These establishments could open the way for effective production of green oil using both diatoms. In addition, these two diatoms could grow under high CO2 concentration, suggesting the possibility of microalgal capturing of exhaust gas from power plant.
