The purpose of our project is to develop a fluorescent microscope to observe in-site nano-carbon materials dispersed in liquid or polymer. Based on a total internal reflection fluorescence microscope (TIRF), we have developed a new microscope using the fluorescence light emitted by the dye dissolved in solution as a background illumination. Using this microscope, we are able to image thin graphene oxide (GO) sheets dispersed in water that are not visible by a bright-field microscope or epi-fluorescence microscope. It is possible to image GO sheets at any wavelengths that are selectable by the dye. Not only GO sheets that are attached to a glass surface, but also sheets that float in solution are confirmed to be observable by the experiments as well as a simulation.
At low dye concentrations, the evanescent field near the glass surface contributes most to the excitation of dye. At high concentrations, a refracted beam becomes dominant which makes GO away from the glass surface visible. In particular, partial transparency of GO sheets allows to observe laying structures at high dye concentrations. This is an unexpected result, as we have thought that GO sheets appear totally dark due to FRET.
The glass surface was modified by phenylethoxysiline or octadeceylethoxysilane. We have found that phenyl modified surface minimizes adhesion of GO sheets in water.
As a prototype, a nanocarbon observing kit consisting of TIRF microscope (Nikon Instrument), a sample dish, a spacer ring, chemically modified glasses, and dyes was made for commercialization.
