 We have cultivated Nicotiana benthamiana under several environmental stress conditions and have obtained profiles of the secondary metabolic-related gene expression to provide information to improve the efficiency of secondary metabolite production in plants. In this study, we established real-time PCR assays for 24 key enzyme genes on the main secondary metabolite pathway. To obtain expression profiles of the genes in question, we extracted total RNAs from N. benthamiana leaves. Subsequently, we synthesized cDNAs from these RNAs and subjected to the gene expression analysis by real-time PCR. Real-time PCR analysis showed that 4 of the genes were not expressed on unstressed N. benthamiana leaves. This year, we have cultivated N. benthamiana with 12 kinds of chemicals and two different light conditions as environmental stress conditions. As a result, it was found that many genes were differentially expressed in response to the 12 kinds of chemicals. For example, real-time PCR analysis showed that the expression levels of 10 of the genes were approximately 5-fold higher in a chemical-treated plant than unstressed plants.
