Neurodegenerative diseases disrupt functions of the nervous system by killing specific types of neurons. Some mechanisms of the neurodegeneration have been shown by recent researches, but they are still unclear. There is no effective drug yet, so the improvement of the drug screening system is necessary. The situation of cultured cells conventionally used for a drug screening has a big difference from in vivo situation. If some compounds were effective for the cultured cells, it’s unknown that the compounds are really effective in our bodies. So it is important to reconstruct the in vivo-like tissues (organoid) and apply the organoids to the drug screening.
Our purpose in the SUI project is to establish a screening system for a drug discovery of neurodegenerative diseases. The system consists of two parts. One is a microfluidic device to reconstruct three dimensional neural tissue (organoid) in parallel and the second one is a soft ware to quantify the level of neural tissue’s degeneration. Until 31th March 2017, we mainly investigated the material for the fabrication of a prototype-microfluidic device. First we planned a fabrication to make a prototype-microfluidic device made by polystyrene (PS), which is generally used for tissue culture-equipment. However, the fabrication of PS prototype device required high-cost and a long lead time. So we shifted the material to Cyclo olefin polymer (COP). It was shown by previous researches (Su, Xiaojing, et al., 2011) that COP was useful for a cellular culture-device, then we confirmed the possibility to reduce-cost and time. From April 2017, we actually tested the COP prototype device for the organoid generation. After final testing of the prototype, we will decide the material for the organoid culture plate.
