英文要約Surveillance study on specific food allergen testing by rapid and simplified quantitative system construction (FY2008-FY2009) FY2008 Annual Report
We aimed at a specific food allergen testing rapid and the simplified system construction, and development is advanced by the three as a theme of the (1) marketing research, the (2) easy pretreatment method and the (3) establishment of the detection technology. Ten sites were visited to grasp the situation and the requirement for the automation of specific allergen testing. As a result, shortening of the sample extraction step and simplified testing without needing special knowledge are requested on the site. It has been understand that the affordable system which the result comes out within one hour after setting sample is hoped for. The pre-preparation described in the notification method is not a method that can be easily introduced into the factory by the food entrepreneur due to the complex operation process. Therefore, it aimed to make establishment of the process that was able to be done handily in a short time and a safe extracted material that treated easily. The prospect that the extracted material for the testing is obtained without using the centrifuge by an original stir tool and an easy preprocessing device in 12 minutes attached. Moreover, it was confirmed to have the same reducing power as the existing reagent to extract a target protein from food sample. The process should be established and then the product development with the performance described to the notification method as a tool for the preprocessing of the ELISA is advanced. The offered antibodies were first selected using ELISA and new metrology which is named BIST (Bead array in single Tip: BIST incorporates beads aligned in a capillary tip for ease of use and the option to perform the system manually with use of a pipette) for multiplex signal detection system. It was confirmed to be adaptable to use BIST for the qualitative measurement of a specific raw material. In addition, the possibility that the fixed quantity value can be presented is possessed. The detection sensitivity of BIST at present was equal to ELISA. In BIST, the simultaneous detection using three purified antigen and the possibility of multiplex detection of the egg and the milk in a cookie were demonstrated. In order to be quantitative assay using BIST, the examination that uses the bead that makes the antigen of numerical density immobilized as an internal standardization is done. Principle prototype of the detection unit was improved by modification of nose geometry of use fiber. Development are advance using suitable antibodies for BIST as a multiplex testing and original system from the preprocessing to the result of a measurement.
