and Biofuel Production

This R&D objective is to develop innovative means for converting renewable
lignocellulosic biomass to practical liquid fuels such as bioethanol and biodiesel. To
achieve this, the R&D implements selected enzymes and genes in improving
saccharification of biomass and breeding of C6/C5 fermenting organisms for bioethanol.
The first fiscal year, four sub-themes as follows were started with 18 members.
Theme 1): To get better understanding of cellulosic materials towards effective
enzymatic saccharification, four resources were selected as starting biomass:
Japanese cedar, eucalyptus, rice straw and Erianthus (an energy crop). A combined
analysis of microstructure of lignocellulosic materials and their interaction with
enzymes is designed to better understand the complicity of mechanism, which is
prerequisite for achieving efficient saccharification.
Theme 2): To obtain potentially useful microorganisms, enzymes ,and genes for
improved biomass saccharification, we conducted R & D taking full advantage of cutting
edge technologies such as metagenomics and transcriptomics as well as high
throughput screening of organisms from culture collections. Attempts were also made to
use the ABI SOLiD DNA sequencer for de novo sequencing
Theme 3): To produce potential enzymes and their utilization towards effective
saccharification, the R & D conducts standardization of determination of activities,
stabilities, and substantial traits of cellulolytic enzymes for their practical use.
Available or newly found enzymes will be assessed to precisely evaluate their enzymatic
potentials under the optimized conditions. This fiscal year, the subcommittee has 13
subjects containing 3 outsourcing, the standardization of reducing sugar determination
and cellulase activity measurement were carried out in JBA Tsukuba laboratory. In
addition cloning and heterogeneous expression of a lot of genes of cellulases,
hemi-cellulases and proteins useful for scarification from various origins using various
host-vector systems will be conducted.
Theme 4): As screening and breeding of fermenting organisms for biofuel and biodiesel
production, this R & D focused on Saccharomyces cerevisiae as an ethanol-producing
platform, and attempts to confer these traits by various means of genetic and breeding
manipulations and construction of platform organisms for biodiesel production.
Furthermore we attempted to obtain genetic information data regarding metabolism
and stress tolerance of microorganism which is useful for fermentation of saccharified
C6 and C5 sugars using next generation sequencer.
